The CarboSep CHO-413 HPLC column (300 × 7.8 mm) is based on a polymeric styrene-divinylbenzene matrix in lithium form. CarboSep CHO-413 has a cross-linking degree of 4 per cent and is used in ligand-exchange chromatography for carbohydrates. The combination of a lithium counterion and a low degree of cross-linking makes the phase particularly suitable for the analysis of oligosaccharides and more complex carbohydrate mixtures. The CarboSep CHO-413 column is typically operated with aqueous eluents and at elevated temperatures to achieve stable retention and good peak shapes.
The CarboSep series uses polymer-based packing materials with defined counterions and degrees of cross-linking. Unlike conventional silica phases, retention is not based on hydrophobic surface interactions, but rather on ligand exchange and size- or structure-dependent interactions between the carbohydrates and the polymer resin. This allows sugars, sugar alcohols, and oligosaccharides to be separated in highly aqueous eluents. The polymer backbone used is designed for routine applications in carbohydrate analysis; compatible CarboSep pre-column cartridges help keep matrix components out of the main column.
