The Capcell Pak C18 MGII HPLC column250x4.6mm, 5µm, bears an octadecyl modification (ODS, RP-18, C18) on polymer-coated silica with 10 nm pores, is assigned to USP class L1 and operates in the pH range 2 to 10. The coating here is further developed so that silanol activity is particularly strongly suppressed, which allows basic analytes to elute symmetrically, especially under neutral conditions, i.e. in the most critical case for bases.It is the first choice for new method development because it favourably combines plate number, tailing factor and backpressure, and method transfer between particle sizes is secured by versions with identical selectivity.Demonstrated are aldehydes derivatised with dinitrophenylhydrazine from formaldehyde to isovaleraldehyde as well as an oligonucleotide length standard, and with well over two hundred application notes this phase has the broadest application base of the brand.

Capcell Pak employs a unique design principle, which the manufacturer refers to as the “capsule type.” The silica support is not silanized directly; instead, it is first coated with a uniform, thin layer of silicone polymer, and the functional groups are bound only to this layer. In this way, the brand solves a fundamental problem in reverse-phase chromatography: Conventional octadecyl phases hydrolyze in alkaline conditions even after end-capping, causing the silica support to dissolve, whereas purely polymeric phases exhibit lower separation efficiency, lower compressive strength, and swelling behavior. The polymer layer therefore replaces end-capping and is the reason for the brand’s wide pH ranges. The silica is also tested for its metal content, which is essential for complex-forming analytes. The individual phases can be plotted on a diagram of hydrophobicity versus surface polarity, which serves as the key to phase selection within the brand.
