The Chiralcel phases are based on cellulose-derived polysaccharides that are physically coated onto a silica scaffold to serve as chiral selectors. The cellulose structure creates an ordered, chiral environment, while the specific derivatization—for example, phenylcarbamate, benzoate, acetate, or cinnamate—determines the specific hydrogen-bond, dipole, π-π, and hydrophobic interactions. The advantage of the coated Chiralcel phases lies in their very broad, proven selectivity and often good loading capacity, particularly for preparative separations. The disadvantage is limited solvent compatibility: highly solvent organic solvents can impair the physically applied polysaccharide layer. For a robust method, Chiralcel pre-column cartridges with a suitable phase should be used, as chiral polysaccharide phases can be sensitive to matrix components, particles, and strongly adsorbing sample components.
