Mixed-mode phases for HPLC utilise different separation techniques in parallel in one column. The stationary phases are specially/differently modified for this purpose so that, for example, reversed phase and ion exchange properties are simultaneously involved in the separation. Nowadays, there are many different mixed-mode phases that combine a wide variety of separation modes. They exhibit unique selectivities, making them the column of choice for some difficult separations.
The selection of the correct mixed mode column depends on the analytes and their ability to enter into certain interactions. For example, if a sample contains anionic and non-polar analytes, a mixed-mode column with anion exchange and reversed phase properties should be selected. Up to four different interaction mechanisms can take place simultaneously in one column, whereby the mobile phase is often decisive for the type of interaction or the strength of the interaction. As the selection depends on the analytes in a sample, it is difficult to establish general selection criteria.
