The Chiralcel OD-I HPLC column 300x4.6mm, 20µm, based on a cellulose tris(3,5-dimethylphenylcarbamate) modification on a 20 µm silica support. Unlike the conventional coated Chiralcel OD, the modification in Chiralcel OD-I is immobilised. The phase is therefore cellulose-based rather than amylose-based. Immobilisation ensures that the chiral selector is more firmly fixed to the silica matrix, enabling significantly broader solvent compatibility than with coated OD phases. At the same time, the selectivity of Chiralcel OD-I may differ from that of the coated Chiralcel OD, so existing methods should be verified chromatographically. Chiralcel OD-I is suitable for chiral HPLC and SFC methods where the OD-like cellulose selectivity is to be combined with greater solvent flexibility, for example, in the case of poorly soluble racemates, preparative separations and scale-up processes.

Chiralpak includes both amylose-based and cellulose-based polysaccharide phases. In the immobilized phases, the chiral selector is fixed to the silica support. This allows for the use of significantly more organic solvents than with coated phases, which is particularly helpful when samples have limited solubility. Coated Chiralpak phases are based on polysaccharide derivatives physically applied to silica. They have a long history of use and often offer very high selectivity and good loading capacity. The most important limitation is solvent compatibility: mobile phases or sample solutions that can dissolve the polysaccharide layer must be avoided. Immobilized and coated variants with similar modifications are not necessarily chromatographically interchangeable. Immobilization increases robustness and solvent flexibility but can alter the spatial accessibility of the selector. Therefore, methods should be reviewed when switching between coated and immobilized phases and reoptimized as needed.
