The Chiralcel OX HPLC column 50x4.6mm, 5µm, based on cellulose as the polysaccharide backbone. The specific modification used is cellulose tris(4-chloro-3-methylphenylcarbamate); the chiral stationary phase is physically coated and applied to spherical silica. Chiralcel OX is primarily used in normal-phase chromatography and SFC, and, depending on the version, also in reverse-phase chromatography. The separation of the enantiomers occurs via hydrogen bonding, dipole-dipole interactions, π-π interactions and steric fit. Typical applications of the Chiralcel OX include aromatic, halogenated and structurally similar enantiomers used in drug discovery and quality control. The alternative positioning of chloro and methyl substituents in Chiralcel OX ensures that it recognises enantiomers in a manner complementary to that of Chiralcel OZ.
The Chiralcel phases are based on cellulose-derived polysaccharides that are physically coated onto a silica scaffold to serve as chiral selectors. The cellulose structure creates an ordered, chiral environment, while the specific derivatization—for example, phenylcarbamate, benzoate, acetate, or cinnamate—determines the specific hydrogen-bond, dipole, π-π, and hydrophobic interactions. The advantage of the coated Chiralcel phases lies in their very broad, proven selectivity and often good loading capacity, particularly for preparative separations. The disadvantage is limited solvent compatibility: highly solvent organic solvents can impair the physically applied polysaccharide layer. For a robust method, Chiralcel pre-column cartridges with a suitable phase should be used, as chiral polysaccharide phases can be sensitive to matrix components, particles, and strongly adsorbing sample components.
